چکیده

Also known as... The terms ‘centromere’ and ‘kinetochore’ are often used synonymously but it is best to consider the centromere as the DNA and chromatin on which the kinetochore is assembled and the kinetochore as the protein machine, including centromeric chromatin, which mediates attachment to the spindle. Centromeres are found near the centre of the chromosome on metacentric chromosomes, but they can also be submetacentric, between the middle and an end; acrocentric, close to an end; and telocentric, at an end. Some organisms such as Caenorhabditis elegans are holocentric and assemble centromeric chromatin and kinetochores along the entire chromosome. Delving further you discover that centromeres are eccentric!

Discovery of an Unconventional Centromere in Budding Yeast Redefines Evolution of Point Centromeres

Centromeres are the chromosomal regions promoting kinetochore assembly for chromosome segregation. In many eukaryotes, the centromere consists of up to mega base pairs of DNA. On such "regional centromeres," kinetochore assembly is mainly defined by epigenetic regulation [1]. By contrast, a clade of budding yeasts (Saccharomycetaceae) has a "point centromere" of 120-200 base pairs of DNA, on wh...

Repeatless and repeat-based centromeres in potato: implications for centromere evolution.

Centromeres in most higher eukaryotes are composed of long arrays of satellite repeats. By contrast, most newly formed centromeres (neocentromeres) do not contain satellite repeats and instead include DNA sequences representative of the genome. An unknown question in centromere evolution is how satellite repeat-based centromeres evolve from neocentromeres. We conducted a genome-wide characteriz...

Maize Centromere Structure and Evolution: Sequence Analysis of Centromeres 2 and 5 Reveals Dynamic Loci Shaped Primarily by Retrotransposons

We describe a comprehensive and general approach for mapping centromeres and present a detailed characterization of two maize centromeres. Centromeres are difficult to map and analyze because they consist primarily of repetitive DNA sequences, which in maize are the tandem satellite repeat CentC and interspersed centromeric retrotransposons of maize (CRM). Centromeres are defined epigenetically...

Gene Expression and Chromatin Modifications Associated with Maize Centromeres

Centromeres are defined by the presence of CENH3, a variant of histone H3. Centromeres in most plant species contain exclusively highly repetitive DNA sequences, which has hindered research on structure and function of centromeric chromatin. Several maize centromeres have been nearly completely sequenced, providing a sequence-based platform for genomic and epigenomic research of plant centromer...

Regional centromeres in the yeast Candida lusitaniae lack pericentromeric heterochromatin.

Point centromeres are specified by a short consensus sequence that seeds kinetochore formation, whereas regional centromeres lack a conserved sequence and instead are epigenetically inherited. Regional centromeres are generally flanked by heterochromatin that ensures high levels of cohesin and promotes faithful chromosome segregation. However, it is not known whether regional centromeres requir...

Nuclear distribution of centromeres during the cell cycle of human diploid fibroblasts.

The distribution of centromeres in the interphase nuclei of human diploid fibroblasts was analyzed using anti-centromere immunofluorescence and laser scanning confocal microscopoy. The positions of the centromeres were placed within the nuclear chromatin distribution and presented some aspects of the dynamics of nuclear structure during the cell cycle. During the G1 phase of the cell cycle many...
